Can anyone tell me what is the best way to get rotor bolt out on a 81
CB750 I'm having a bit of trouble and wanted to ask someone before I
mess something up...?

Alternator rotor:

It might be possible to pull the alt rotor by using a gear puller on
the pole pieces and leaving the nut real loose to pull against, but
the halves of the pole pieces may come apart before the rotor comes
off - not that bad if your intent was to take it apart anyway.

The shaft is tapered and the pole pieces have a taper so a little(!)
tapping with a hammer might help.

The two halves of the alt rotor are only held on by four peened areas
over the joints.  To separate them I used a gear puller.  The slip
rings are held on by four machine screws cleverly buried under some
black epoxy or something - that has to be removed before you can see
the screws.

Yup, that's why leaving the bolt in makes sense - assuming he's going
to try to do it without buying the puller.  It may be the same puller
that was used on the single cam 750 - I know the same puller works for
the (external) rotor on the VF 1100 S

There was no way to get any of the gear pullers I owned to grab the
base of the rotor, but they could grab the pole piece - and risk
pulling the halves apart without budging the base.  But it did look
like the gear puller might work once the pole piece was split.

If the rotor is shorted anyway, there's nothing lost by doing it.  If
the rotor is good, that would break it.
